While most people would be upset at their loved one falling out with them so bad that they're willing to maim them, the Joker's issue with this is far different. Given that the Joker considers himself Harley's creator, her turning on him so violently likely aggravates the clown further because it shows he can no longer control her as he did.

Also, it likely messes up his whole 'scary evil smile' shtick, which is also sure to have wound up the criminal jester yet further.
