Newt isn't going to be on his adventures alone, oh no. Second billed in the film is Katherine Waterston as Porpentina Goldstein. Judging by the trailer, she'll team up with Mr. Scamander early on in his time in America and will serve as something of a guide (at first at least).

Little is known about her, although we have learnt from set reports she works for the Magical Congress of the United States of America (the US version of the Ministry Of Magic) and has run afoul of superiors standing up for the little man, so expect a 'strong female character' here.
