1. Thanos Seemingly Gets The Fully Powered Gauntlet

The end of the D23 footage showed Thanos with the Gauntlet, with some reports that it was fully powered for that loaded shot. If that's true, it's a massive, massive deal, because it spells disaster for a lot of characters. There's no way they can fight Thanos with the full Gauntlet and no way they could hope to even dent his plans.

Other footage descriptions suggest that Thanos is seen wearing the Gauntlet with only two Stones in (blue and purple), for the off-planet battle with Iron Man, Doctor Strange and the Guardians, and that's enough to literally let him pull a planetoid (or moon) out of the sky to throw at his enemies...

So there's a pretty big indication of why it's bad news for him to get his hands on all of them. That interestingly also suggests that Xandar will fall, since that's the current location of the purple (ie Power) stone after the Guardians put it there for safe keeping in Vol 1.

And if you're looking for further indication that he gets all of the Stones, the concept art above that heavily channels the cover of the Infinity Gauntlet comic has all of them there, glistening ominously.
