In the comic book Infinity Saga, Thanos' quest to unite the Infinity Gems is very much tied to his desire to impress and get under the robes of Lady Death. The infamous click of his fingers that wipes out half of existence is basically an attempt at foreplay, in other words.

So far, we don't have any sort of idea of Thanos' motivations. Maybe he just wants to be powerful above all else, but then he's already been called the most powerful being in the universe (even if he's done precious little to deserve the billing thus far), so it seems like overkill to seek even more power. We can assume then that he's either just massively arrogant or he has a point to prove.

If it's the latter, it could still be that he is trying to woo Death, though that might be a supernatural character too far at this stage of the MCU. Perhaps the Russos will instead go with a supremely powerful character associated with death who is already just about to appear in Phase 3... Cate Blanchett's Hela. It wouldn't be too much of a change for Thanos to be besotted with her rather than Death, and that wouldn't require much change to the comic source.
