Though the path to Thanos getting his hands on all of the Infinity Stones remains unset so far, we know that he's going to get his hands on at least a couple pretty quickly, which will exponentially increase his power and the threat he then poses to the Avengers and Earth. The rest of the Stones should follow, and that spells disaster.

And there's no way he isn't going to have his moment to show what the fully empowered Gauntlet can do, and it would be foolish of the Russos not to reference the infamous panel in the Infinity Sage when the Titan clicks and wipes out half of existence.

If there's any sense to the movie, that will happen right at the end, setting up Avengers 4, which will see the heroes seeking their redemption, and it would be the bravest way to end a comic book movie ever. Imagine the impact it would have on the fans, for a start.
