It's been clear to audiences for some time now, but the heroes of this movie are going to be discovering just what Thanos wants: the Infinity Stones. As Tony Stark says: "We have what Thanos wants, so that's what we use."

That includes the Time Stone, which is currently in possession of Doctor Strange, and the Mind Stone, which is unfortunately embedded in Vision's skull. It's no surprise that he looks worried.
