At the minute, it's a little unclear whether Gotham City Sirens is actually still a separate project from Birds Of Prey. The fact that both would feature female metahuman teams led by Harley Quinn (apparently) makes co-existence unlikely. But as late as late 2017, Robbie was talking about hiring the right director.

It just looks like Warner Bros might have quietly shelved it to focus on other things for now.

Will It Happen?

It should, because of the potential of the characters, but having yet another movie for Harley Quinn and The Joker to dominate (whent here are better characters to focus on) would be a mistake. And it might well be that Warner Bros decide to focus on Birds Of Prey and the Joker's movie separately first.

The fact that none of the updates on Birds Of Prey have mentioned GCS seems a little ominous too.
