6. Drax
The trend for more recent MCU movies has been for the villain to have a more personal link to the hero and for that personal link to matter at the end of things. The revelation of Zemo's relationship with the Avengers was incredibly effective and so too was the closeness of Erik Killmonger and T'Challa in Black Panther.
And while Thanos' plot might have universal ramifications, his story is tied to some of the Avengers in a very personal way that could be used to plot his downfall. Obviously, he has two "children" aligned with the heroes, but he also has a walking, talking angel of vengeance to deal with too in the shape of Drax The Destroyer.
In the comics, Drax was created solely to destroy Thanos and while that element of his backstory is missing from the movies, the drive - of his murdered family - remains. Crucially, so too does the hunger to kill him.
And while he's something of a comic character, Drax is hugely powerful and he legitimately could go toe-to-toe with the Titan. Him succeeding would be particularly fitting since Thanos' actions seem to suggest he has no considerations for accountability. Death by irony, if you will.
