While fans have seen Spider-Man clean up the streets and, more often than not, the sewers, of New York City countless times before, it could be time for the hero to be put into more surreal, otherworldly contexts.

Mr. Negative as the villain already opens up the possibility for the developers to pull the rug out from under players, but the inclusion of other tricksters like Mysterio could facilitate imaginative sequences that don't take place in the real world. Likewise, if they really wanted to go all out, the studio could include some cosmic threats that literally transport players to out-of-this-world environments.

Moving the Web Head out of his comfort zone and into more fantastical settings - whether they're real or not - will give the developers the freedom to explore this character in ways fans haven't seen before in a video game, and quickly distinguish this release from ones that have come before.
