As for who plays the riff, watching YouTube clips it seems that it was interchangeable, with Lennon often playing the intro version but sometimes letting Harrison play it whilst Lennon was singing.

The secret is in maintaining a strong barre across the neck and ensuring you've got the pinkie strength to hop between the 3rd and fourth strings. Master it at the 10th fret before building up to the full pattern including a much tougher stretch at the third fret.
