Although Masters is very much a scientific man, and also a married one, that doesn’t stop him from developing feelings towards Virginia. The relationship between the pair developed from scientist and secretary to partners in research, and as part of that they began to have sex as well.

At first it’s purely just to aid the study, but it becomes something more, certainly from Masters’ point-of-view but also Johnson’s as well, and there’s a lot of sexual tension and stolen glances between them. 

Similar to how Masters was changed for TV, this seems to be something played up to add a bit more excitement and drama to the story, as they were supposedly very formal and professional in the lab, described as being “aloof” with each other.
