It'd almost be a shame for The Joker to play that big of a part in the film, anyway, since his appearance might overshadow another supervillain with a taste for the theatric and comical. Harley Quinn might have been brought in as The Joker's girlfriend (having been a big hit in the Batman Animated Series, no less), but she's since become so much more than that. Owing to her popularity in the Arkham game series, she's recently been spun off into her own phenomenally popular solo series and duh the current line-up of the Suicide Squad. Dr Harleen Quinzel was The Joker's therapist at Arkham Asylum, who got lured into a twisted love affair with the villain, which may or may not be felt both was. Anyway, she's gotten way more interesting than that. Whilst she began as The Joker's Girlfriend, Harley Quinn, putting the character in a solo book and having her operate as a member of the Suicide Squad has allowed her to grow. At least, a little. She's still fairly Joker-obsessed, but her ruthless efficiency in the field (possibly owing to the fact the government put a bomb in her to make sure she cooperated) has made her into a sort of defacto team leader alongside Deadshot. If the plot of Assault On Arkham is informing Suicide Squad, she'll be brought along thanks to her familiarity with the facility, and with The Joker. Either way she's going to be more than a little deranged and high energy, in which case Margot Robbie who broke through with a brilliant turn in The Wolf Of Wall Street is another pitch perfect bit of casting.
