6. Thanos Gets The Infinity Gauntlet (Avengers 3)

We know the Infinity Gauntlet exists in the Marvel Cinematic Universe - we saw it, albeit very briefly, in the first Thor movie inside Odin's trophy room - and we know that the Infinity Stones (the Marvel Cinematic Universe versions of the Infinity Gems) exist in the same universe, as we've already seen the Tesseract, the Aether and the Orb. Moreover, we know that Thanos wants them. Given how iconic the Infinity Gauntlet comic book arc is, that first moment Thanos slips on the completed gauntlet will indicate the start of a threat to the entire Marvel Cinematic Universe on a level far, far, far above anything we've seen so far. Hopefully, the movie will capture the essence of the moment Thanos first got his hands on the gauntlet in the comic books, as it will be one very awesome moment.
