In the comic books, S.H.I.E.L.D. sought out Blade in an attempt to use his blood for Project: Silvereye - an attempt at cloning vampire operatives for their agency. It could make a very cool movie arc if S.H.I.E.L.D. learn of Blade's existence and aggressively hunt him down in an attempt to capture him for this very purpose. The movie wouldn't need to adhere to the comic book plot elements - as, frankly, it wasn't a great or huge story - but it would allow for the likes of Hawkeye, Black Widow, Melinda May et al to have further Marvel Cinematic Universe roles in hunting down the vampire hunter and would be exciting throughout on the big screen. Ultimately, like in the comics, Blade could shut down Project: Silvereye and form an alliance with S.H.I.E.L.D., working in their supernatural division alongside the likes of Doctor Strange. The organisation could later supply him with new equipment - including his gun-hand in the event that he has to chew his own hand off in the rebooted version of the character's adventures - and send him out on missions in the same way they do with their current agents. Essentially, S.H.I.E.L.D. initially being Blade's adversaries, chasing him down, and Blade ultimately joining S.H.I.E.L.D. simply oozes potential awesomeness and we would absolutely love to see it happen.
