It's never been suggested that Stephen Strange might be gay in the comics, but then he's never really had any kind of intimate relationships of note. For the most part he seems to be depicted as kind of asexual, choosing to focus all of his time and energy on mastering those mystic arts. So since the Strange is in no way defined by his sexuality, why not introduce him as Marvel's first openly gay character - one who'll be headlining his own movie, no less. 

It's really not that much of a stretch anyway: Strange is a learned, neat, well dressed man in his mid-thirties who lives with 2 male room-mates in an apartment furnished with antiques. Plus, he even has a mustache!

All joking aside, this would probably rub a few fans the wrong way, but so what. If Marvel are serious about giving an LGBT character the spotlight, then they should really go for it and put their name on the marquee. Plus, you just know Benedict Cumberbatch would do wonders with the subtle nods to Strange's homosexuality.
