For those who are unfamiliar with the source material, Avengers: Infinity War is based on a six-issue comic-book series titled "The Infinity Gauntlet."

The story details Thanos' (played by Josh Brolin in the MCU) efforts to mount the six Infinity Gems on a glove to form the Infinity Gauntlet, granting the character omnipotence.

With the Gauntlet assembled, Thanos seeks to win the affections of Death - or rather, the physical embodiment of death. In order to do so, Thanos wipes out half the sentient life in the universe with a mere click of his fingers.

Just as an aside, you should never do that on the first date. A movie and a meal will be fine.

So far in the MCU, we've seen five Infinity Gems (or Infinity Stones, as they're called in the movies).

The Space Stone (the Tesseract/blue cube from The Avengers), The Mind Stone (the yellow object residing in Vision's forehead that came from Loki's scepter), The Power Stone (the purple orb from Guardians Of The Galaxy), The Reality Stone (The Aether from Thor: The Dark World) and The Time Stone (the green Eye Of Agamotto from Doctor Strange).

The final one that's yet to appear is The Soul Stone. Some fans think it resides inside Thor's hammer or even inside Heimdall, but its true location has yet to be revealed.

The connection between the movie and the comic goes deeper than just The Infinity Stones, though. Speaking with Collider, Marvel Studios president Kevin Feige had this to say about Thanos' plan:

“[His motives] will be similar to what they were in the Infinity Gauntlet in various ways. I guess that’s all I can say about that.”

That could mean Thanos will be chasing after Death in the movie, just like he did in the comic-book. His first appearance at the end of The Avengers, and the line about courting death, does seem to heavily indicate that this will be the case.
