Iron Man now seems like an obvious choice to kick-off the MCU, but in reality it was only done somewhat out of necessity. Marvel had sold the rights to its biggest heroes off back in the late nineties due to bankruptcy, meaning that by the time the opportunity to set up their own movie studio came around they had to do it with their B-List heroes, who didn't have the mainstream appeal of Spider-Man or X-Men. As a gamble in its own right, let alone in terms of what it was leading to, there's not much in the way of genuine foreshadowing present in the first Iron Man (there could easily have been no follow-ons). Although that doesn't mean there aren't a few cheeky winks to other heroes (some of which have been ret-conned to have big significance). And that's saying nothing of the film's attention to comic detail; there's numerous specific references to the character's past, in print or otherwise, that go to show how carefully constructed the film was.
