For a lot of fans, Supernatural ended with Season 5. While that was in part because it was the end of original creator Eric Kripke's run, it was also because its finale, Swan Song, capped off the main storyline in such an elegant way. Paying homage to the past five years, the climactic episode was essentially a love letter to Sam and Dean's relationship, with it being their bond - not any spell, weapon or trinket - that ended up saving the world.

Watching these two regular guys (well, regularish) go up against the strongest warriors both Heaven and Hell had to offer paid off pretty much every question the show posited about free will vs. determinism. It helped that it was a bloodbath as well, with Bobby, Castiel, and even Sam all being killed off (albeit temporarily) before the episode was over.

Consequently, it's undermined a touch from having to undo some of its larger narrative moments to continue the story in Season 6, but even in spite of that, Swan Song offered the perfect end to one of the best horror shows on TV.
