1. Death Is Her Gift
Any of the moments we have discussed could be number one, but Buffy's death (technically, her second) at the end of The Gift is such an emotional scene that you would be hard-pressed not to pick it as the best the show can offer.
Again, the image of Buffy running into the portal to save her sister, her friends and the whole planet is such a definitive one for her character. Her final speech is one of the character's best, too. "The hardest thing in this world is to live in it," she tells Dawn. "Be brave. Live. For me."
I'm glad this wasn't the end of the show, but it would have been a terrific closer if it had turned out to be. That final shot, showing the epitaph on Buffy's gravestone, is the bit that really raises a lump in your throat: "She saved the world. A lot."
