5. Ultimate Spider-Woman
The Ultimate Jessica Drew is not just a clone of Peter Parker, she also has all his memories up to that point in his life. In other words, she is basically Peter Parker in a genetically modified female body. She has all the same powers as Spider-Man and she retains his attraction to women.
Not only does she become an effective superhero in her own right, but she becomes an agent of S.H.I.E.L.D. and eventually joins the Ultimates, this world’s version of the Avengers. All of that means she could pop up in either the Avengers or Agents of Shield, and she could work best as a new liaison between the two because the use of her powers could be reproduced inexpensively on the smaller screen.
Marvel’s Spider-Man is all set to make his official debut in the Marvel Cinematic world soon and more of his solo movies will follow so he’s going to need a supporting cast.
Marvel has shown an affinity for adapting stories straight from the comics for their movies and there is plenty of material from Spider-Man’s various clone sagas. Of course, Spider-Man and clones are a touchy subject in the comic book world because of the way certain stories were handled in the past but that doesn’t mean it can’t be done well.

Olivia Thirlby from Dredd and Juno could be a good choice to play Jessica.
