Delivering the biggest gut-punch from all the films in the MCU is, ironically, the opening scene from their most popular comedy.

With his mother dying of brain cancer as he listens to the "awesome mix" that she gave him, young Peter Quill is brought into the hospital room by his grandfather to visit her. Refusing to look at his mother deteriorated almost to nothing, Peter receives a wrapped gift from her as she bids him to open it when she's gone.

Seemingly denying her words, angry at the reminder that he's going to lose her, Peter turns away as she asks him to take her hand. She begs him to take her hand, and in the moments in which he refuses to do it, she flatlines and passes away, with Peter immediately turning to her and screaming denial and regret for missing his last chance to hold his mother alive.

Sadder still is his grandfather bidding him to stay right where he is, only for him to be abducted by an alien ship as he runs outside the doors. For being one of Marvel's funniest films, Guardians of the Galaxy certainly has the most painful, heartbreaking moment in all of the MCU's filmography.
