Felicia Hardy was actually introduced in Marc Webb's Amazing Spider-Man 2 as played by Felicity Jones, but they didn't bother giving her much to do and seemed to be setting her up for potential sequels (of which there will thankfully be none), so we have yet to properly see Black Cat in all her glory on screen.

Now that the Marvel/Sony deal allows for access to Spider-Man's supporting cast, there's a great opportunity to reintroduce the character as a lesbian in the Jon Watts-directed reboot. This wouldn't really even be considered altering her sexuality, as the Earth-982 incarnation of Hardy is gay.

As she is so often depicted in the comics, Hardy could start out as a villain before teaming up with Spidey to tackle a larger threat. There doesn't have to be a big song and dance made of her lesbianism either - a single scene of her flirting with Mary Jane (or Gwen Stacy, but after the Webb movies that doesn't seem likely!) or a photo of a girlfriend on a desk and it's established.
