The Scene

After Ultron's defeat, we get a strange shot within what appears to be a vault as it opens to reveal the Infinity Gauntlet. Thanos puts it on and says "fine, I'll do it myself".

What It Meant

Infinity War is coming, in short.

The post-credits is something of a lie, because it suggests that he's been sending minions out to get ahold of the Infinity Stones and they've failed each time. That was the case with Loki (but Thanos deserves to be shot for giving him an Infinity Stone he then managed to lose) and Gamora, but Ultron was in no way affiliated with Thanos and the Collector clearly didn't hand over the Aether to him. So quite why he's suddenly got exasperated by the failure of his minions is unclear.

Still, this was the confirmation we knew was coming, that the next big event in the MCU would see Thanos heading to the MCU to get his hands on the Stones.

The scene also introduced another plot-hole, since there was a second Infinity Gauntlet shown in Asgard's vault in Thor. Apparently both do exist, but only one is real.
