There are tons of fantastic comedy pairings on Parks and Recreation, largely because all of the actors have wonderful chemistry with one another, but also due to the fact that they clearly went to great lengths to create unique, genuine relationships between the characters. One of the most interesting has to be Ron Swanson and his devious, sex-crazed librarian ex-wife Tammy II (his first wife was also named Tammy...so was his mother), played by Nick Offerman and Megan Mullally. They have such a strange yet hilarious relationship together, where Tammy actively tries to ruin Ron's life while Ron is alternately attracted and repulsed by her.
