Amara's existence, alongside God’s, is what holds the very fabric of the universe together. We also learn that the Archangels were created specifically to wage war against her and that only with God himself, as well as this army he created, could defeat Amara, aka The Darkness, and seal her away.

Many powerful characters - from Crowley, to Lucifer, to God - all take a shot at Amara and fail, God even being severely wounded in his attempt. Obviously, it goes without saying that a character in the Supernatural universe powerful enough to take down God is a character you don’t want to be messing with.
