Rey Mysterio just had no luck with world titles. In the PG Era alone he held the World Heavyweight belt for a month before getting into that arduous feud with Kane, and the year after that he won a tournament crowning a new WWE Champion, made possible due to CM Punk taking the title hostage. Later that night, before Rey had even had the chance to celebrate winning his first WWE Championship, John Cena sauntered into view.

Cena, the good guy that he is, decided to take that troublesome strap away from Rey in the main event of that same night's Raw, to save Mysterio the annoyance of carrying the thing around airports and hotels. What the creative team was trying to accomplish with that decision remains unknown to this day; it certainly didn't work in getting Cena cheered by the crowd.

It's also worth noting that when Mysterio held the big gold belt, the word 'Heavyweight' was removed from World Heavyweight Championship while he had it. There's probably nothing to read into there, definitely can't have anything to do with the booking of the luchador any time he picked up a world title...
