Leto has made no secret of his physical transformation into the character, having used social media to show the progress made in altering his hair and eyebrows in preparation for the role, so it does make sense that this would be another attempt by him to publicly show his transformation into the character. It came during a gig with his band - 30 Seconds to Mars - when he told the attending fans that the next song would make them get "really crazy" in a Joker-esque growl (the use of the word "crazy" only serving to reinforce the theory that it was his Joker voice).
