1. Aunt May Dies
It's a Spider-Man tradition to lose Uncle Ben, but Tom Holland's Peter has to deal with the death of his beloved Aunt May as well. After Osborne's "darker side" awakens again at the prospect of his being cured, he unleashes an attack on Happy's home, along with the other villains. May stabs Osborne with the cure, but it doesn't work, much to Osborne's amusement. He tosses a pumpkin bomb at Peter and May before flying off, laughing maniacally.
Peter sees an injured May, and has one final conversation with her. He expresses regret at trying to save these villains, but May reminds him that "with great power there must also come great responsibility." Given that this is traditionally Uncle Ben's mantra, saying it aloud practically signs May's death warrant. Shortly after delivering the line, May tells Peter she needs to "catch her breath" as she dies.
We've seen Spider-Man lose his Uncle Ben on screen. We've seen him lose his girlfriend. We've seen him lose his best friend. We've even seen him die himself. But Aunt May always seemed untouchable in the films. Marisa Tomei's version of Aunt May always stood for helping others, and believed in its importance so strongly that it got her killed.
Tom Spider-Man, and the MCU as a whole, will not be the same without Aunt May.
