5. Doctor Hurt
While introducing Doctor Hurt does run the risk of having to include two secret Gotham cults in one game - given Hurt's leadership role within the exclusive and elusive Black Glove society - it seems a sacrifice that'd be more than worth it if pulled off well.
Hurt is the main antagonist in Batman R.I.P - a focal point of Grant Morrison's long-running stint on Batman - and he and the Black Glove go about summarily ruining Batman's life as quickly and effective as you'd imagine a lunatic and his merry band of men would.
Although it doesn't sound particularly special, some of the elements of it - such as the 'Zur-En-Arrh' graffiti that ends up playing a surprisingly important role in the overall story - are actually really unique to the comic itself.
More importantly, it would also work well as a major or minor storyline, meaning that the team behind the game would be free to include it in whatever capacity that wanted, which is especially helpful in filling any gaps in the game.
