For the longest time it made sense that you could only play as a human in Mass Effect. The original trilogy was tied so deeply to humanity's struggle to gain a foothold in intergalactic business that the story simply wouldn't have worked if you could choose to be a different species.

That story is well and truly over however, and there's no reason why multiple playable races shouldn't be an option in the future. Especially if Bioware is going to keep the existing species as the focus of the franchise, it makes sense to let fans finally play as them.

It would bring a fresh perspective to the series for the first time in a while, and allow fans to gain an insight into these different alien races that they could only ever observe from the outside in previous games.

Plus, it would add an intriguing extra role-playing element to the gameplay. Different species have different social customs, and learning to navigate these cultures to get what you want could add a dynamism to the gameplay not present in previous titles.
