One of the many great things about Jessica Jones' first season was its villain, Kilgrave. Played with a charismatic malevolence by David Tennant, he's going to be difficult to top in Season 2 - since, y'know, he definitely won't be coming back.

However, Season 1 did also lay the groundwork for its next villain, in the form of Will Simpson (Will Traval). He started his journey towards villainy towards the end, succumbing to the strength-enhancing drugs, and that put him on a path towards becoming comic counterpart Nuke.

Traval has hinted at his involvement in the second season, and it seems like a given at this point. That means we should almost certainly be seeing Nuke as a major antagonist, though whether or not he paints his face remains to be seen (though you'd expect they would reference it in some way).

Showrunner Melissa Rosenberg has previously said that there might be multiple villains in Season 2, which really just strengthens the case for Nuke's appearance.
