While many people likely thought Kirk would be in the top two, this author would argue DeForest Kelley's take on the ship's doctor remains unique in the franchise to this day, and there's never been anyone like him in the sickbay ever since.

If Spock sits on the logical side of Kirk's mind, then Leonard McCoy is the emotional counterpart, constantly urging his captain to do the right thing in times of peril, weighing on his conscience. His gruffness and dry humour, along with an underlying current of warmth, make Bones who he is - someone who is constantly looking out for the welfare of the crew and will do anything to protect his captain. That's most notable in 'Amok Time', when he is able to prevent Spock from killing his friend by tricking the Vulcan.

Of course, he has his comedy moments too, mainly when he's arguing with Spock about his logical side, and he's become famous within the franchise as the crew member terrified of technology, especially the transporter. He really was one of a kind.
