Every hero has a backstory, and Spidey's is one of the most famous.
Raised by his Aunt May and Uncle Ben, Peter was a normal kid until he was bitten by a radioactive spider, granting him superpowers. Originally he used these powers for profit as a wrestler, until his failure to prevent a crime committed against someone who wronged him resulted in the death of his beloved Uncle Ben. Inspired by Ben's words - "That with great power, must also come great responsibility" - Peter Parker became Spider-Man.
A lot of the time, a superhero's backstory is just an excuse. With Spidey, it's an integral part of why he is the way he is, and informs everything from his double life to his very personality.
He's not a superhero because he just happened to get superpowers - he's a superhero because he wants to be the person his adoptive parents believe he can be, and to make up for the fact that his actions directly resulted in the death of Uncle Ben.
