Although the Green Goblin is most often considered Spidey's arch enemy, others have come close. Doctor Octopus, for instance, was the man who actually invented the Sinister Six. If this is the first time they're assembling, then it would make sense to have him along for the ride.
Doc Ock's inclusion may or may not have already been spoiled by a voice actor for the title, but still, for the time being we don't know if he's actually even in the game. If he is, then it makes sense for him to be the one leading the six, as Otto Octavius is a familiar face to both fans of Spider-Man and to Peter Parker more importantly.
Fans have also claimed that you can hear his tentacles retracting during the footage, but still, if the good doctor is to lead this particular group of villains, there are likely to be few complaints. He's one of Spidey's best villains, and with Spider-Man 2 having obviously spotlighted the character before, he's due a comeback in gaming form.
