The Warhammer 40k universe has expansive lore that has been established and expanded upon for decades. To date only straight to home video and fan made films have been released, the reception of which has varied. That doesn't mean that there's no published material worthy of being made into a film.

Given Hollywood level funding, and perhaps more importantly the go ahead from Games Workshop, incredible movies could be made about the 40k universe. There's enough material for an expansive cinematic universe to be created, spin-offs and all. Action movies, horror movies, tense thrillers, the possibilities for all of these are present in the 40k universe.

All that's needed is for Hollywood to acknowledge the possibilities that are present in the grim dark future for us to receive what every 40k fan has been waiting decades for, to see the best 41st millenium out there up on the big screen.

Macragge, home world of the poster boy Space Marines for Warhammer 40k; the Ultramarines. If the 40k film franchise were to start anywhere it would be here.

The Tyranids, most alien of the many Xenos present in the 40k universe have invaded the home world and system of the Ultramarines. If you have even a passing knowledge of Warhammer 40k you know of the Ultramarines. This story has it all including space combat climaxing in the noble sacrifice of the Emperor-class Battleship Dominus Astra. It detonated its warp drives creating a warp vortex that consumed it and the majority of the Tyranid Hive Fleet.

Drop planet side and initially the Space Marines and their human Auxillia are being overwhelmed. After the Hive Fleet has been destroyed in orbit around a different planet in the system the war was essentially won but would Macragge be lost? When the space-bound Ultramarines returned to Macragge they found few survivors. Even the entire 1st company, one comprised entirely of veterans, had been slain to a man.

This is a perfect "all is lost/Thanos manages to snap" moment. Picture it, one of the few human survivors stumbles out of a wrecked building and looks up to see the decending dropships of the Ultramarines. How can you not want to see that?
