1. Knight Artorias

Knight Artorias is much the same as Manus. His design and lore are great. He is aggressive and difficult to no end. In these regards it would be down to personal preference who took the top spot, but Artorias has the impact, thanks to an emotional weight that Manus doesn't.

Pre-fight, you'll constantly hear about how great Artorias is, and how he is this vigil for all things good. And then you find him succumbing to the abyss. The fight itself is like putting him out of his misery, and that is more impactful than it seems.

Plus, his relation to Sif adds layers to the fight that make not only Artorias one of the best bosses in game, but also in all of gaming.
