2. Spider-Man
The origin story for Spider-Man is one of the most well known in and out of comic book fandom, and it's been left mostly untouched for decades. Peter Parker gets bitten by a radioactive spider, he gains superhuman abilities he uses to earn some quick cash, and his arrogance results in the death of his Uncle Ben.
Pretty much everyone knows this, and it seemed as if his origin would remain without any changes forever, but then J. Michael Straczynski came along and made a tweak that screwed up the whole story. Peter wasn't randomly bitten by a spider, he was chosen by a spider totem-spirit!
When Spidey met up with Ezekiel, he asked him the question of whether he would have gained spider superpowers had the spider bitten him without the radiation. It turned out, he would have, and all the years where Peter had to deal with radioactive blood may not have been necessary.
The totem-spirit chose Peter because he was bullied all the time, and the spirit saw in him an avatar, who, like a spider, would hunt down enemies and serve them some serious social justice. The tweak suggested the bite gave Peter powers, and they were altered slightly as a result of the radiation, but he would have gotten the powers one way or the other.
