6. Spider-Man
The Punisher has gone up against Spider-Man numerous times over the years, and he even made his first appearance in The Amazing Spider-Man #129. In that book, the Punisher was tricked into going after Spider-Man, whom he believed to be the menace J. Jonah Jameson claimed, but by the end of the book, both vigilantes walked away.
Since that debut issue, the Punisher has managed to go after Spider-Man, every once in a while, and on various alternate Earths, he's killed him. A lot. One such killing took place on Earth-95126, which is the Punisher Kills the Marvel Universe reality.
In this reality, the Punisher's family was killed during a superhero battle, so he opted to punish all superpowered people, which of course, includes everyone's Friendly Neighborhood Spider-Man. He gets his chance to take out the wall-crawler when he stumbles upon a fight between Spider-Man and Venom in the sewers.
Frank had previously rigged the area with tasers, which went off, disabling both Venom and Spider-Man, leaving them helpless. Frank first wastes Eddie Brock, taking Venom out of the equation, and then he turns his gun on Spider-Man, who asked why he was being targeted. Frank simply replied, "Because somebody had to be first."
