In The Comics:
In the end, Thanos is betrayed by Mephisto and Death and when he believes he has overcome all of his enemies, he separates his consciousness from his body and assumes an astral form, in effect weakening himself.
At that point his granddaughter Nebula takes the opportunity to steal the Infinity Gauntlet, which he carelessly left back with his physical body. She banishes him and assumes power for herself, replacing him as the big issue for the surviving heroes, who actually end up recruiting Thanos to help.
Eventually, she's defeated by Adam Warlock, who keeps the Gauntlet for himself initially (despite the complaints of the other heroes). He also visits a supposedly dead Thanos on a quiet planet where he's a farmer who has vowed to live in peace (this does not last). Later, Warlock is shown that his desire to "fix" the universe is misguided and is ruled unworthy of the Gauntlet by the cosmic entities of the universe, so he sets up the Infinity Watch to each take a Stone to protect.
In The Film:
This is definitely not how it's going to go down. While we might see Nebula clash with Thanos, it's unlikely he'll go astral and she'll steal the Gauntlet. It just no longer fits her arc. And without Adam Warlock there to solve things, we're getting a completely different ending, clearly.
Whether we'll see the establishment of an Infinity Watch to each keep one of the Stones safe remains to be seen, but it feels more likely that they'll simply be disappeared somehow so Phase 4 can move on to a new over-arching narrative.
