3. Malus (Shadow Of The Colossus)

In the case of Malus, it wasn't so much that he was brimming with personality or earned a lot of our hate, but simply that he marked the end of an epic, exhausting journey, as the sixteenth and final colossus you'd have to take on. Adding to the motivation to take him down is that, on the way to confront Malus, your trusty steed Agro falls, seemingly to his death (though as later revealed, he survives), making the quest that little bit more personal. Malus is a gigantic tower that flings fireballs at you, and even getting to him is a pretty daunting task in of itself. Upon getting to him, you have to climb him in order to reach the sigils that will bring him down, and plunging your sword into those points is extremely satisfying given everything that had led to this point. You would avenge Agro and get your love back, right? Defeating Malus eventually results in a few twists and turns that detract considerably from the satisfaction of killing him (the revelation that killing the Colossi was not such a good idea), but in that moment, the sense of accomplishment was undeniable.
