3. The Hulk
Who hasn't wanted to play as the Hulk in a proper superhero game? The sheer impact of leaping around as the big green guy would make for totally satisfying gameplay. Sure, it's a bit of a shift away from Spider-Man's lithe, athletic combat, but that shift pales in comparison to the enjoyment of being put in Bruce Banner's ripped purple shorts.
Combat would definitively be the main focus of this game; boss encounters would be giant slug-fests, and your everyday thug encounter would more likely be the military trying to prevent collateral damage. Think back to the tank-throwing, ground pounding chaos of Ultimate Destruction, and you're sure to have destructive fun at every turn.
Given Insomniac did their best to focus on the Peter Parker side of Spidey, we could easily tease out more of Bruce Banner's character here as well. Those science mini-games wouldn't be too distinct in a laboratory that Dr. Banner might find himself in.
Hulk has a decent amount of his own storylines to explore (Planet Hulk being a likely contender), so if Insomniac wanted to drastically change styles then The Hulk would be a perfect way to start, now that they've mastered Spider-Man.
